STUDIES ON TUMOR-SPECIFIC CELLULAR-IMMUNE RESPONSES WITH SOLUBLE TUMOR ANTIGEN
Part 11. Kinetics of the Cellular-Immune Responses in Mice with Growing Tumors
Measurements of cell-mediated immunity, specific and non-specific, are important in estimating prognosis and clinical state of cancer patients. In order to see what kinds of assays for cell-mediated immunity reflex immunological states of cancer patients, kinetics of the cellular immune responses in C3H/He mice with growing MH134 tumor, were carried out by the footpad assay, lymphocyte blastogenesis to soluble tumor antigen (MLTR), leukocyte migration inhibition test (LMIT) using soluble tumor antigen and lymphocyte blastgenesis to Phytohemagglutinin (PHA) and Concanavalin A (Con-A).
Soluble tumor antigens were prepared by hypotonic saline extraction method from MH134 cells. These assays were carried out 0, 6, 8, 10, 13, 16 and 21 days after subcutaneous transplantation of MH134 cells.
Responses of footpad assay, MLTR and LMIT reached the maximum 10 days after tumor transplantation, and then decreased along with progressive tumor growth. Lymphocyte-blastgenic response to PHA reached a peek on day 6, Con-A on day 8, and then both declined thereafter. Correlation between tumor diameter and PHA-response was observed. An interesting significant relationship between in vivo reaction of footpad assay and in vitro responses of lymphocyte blastogenesis to tumor antigen and Con-A was demonstrated.
These results suggested the possibility that the measurement of immunological status with the use of soluble tumor antigen might be useful for the study of specific immune responses in cancer patients.
