PROTECTION OF EXPERIMENTAL STRESS ULCERS BY INHIBITION OF COAGULATION, FIBRINOLYSIS AND KININ SYSTEMS

Effect of an anticoagulant, antifibrinolytic agents and kallikrein inhibitors on stress ulcers induced by cold and restraint in rats were studied. Intravenous administration of 50 U /Kg of heparin, 500 mg/ Kg of ε-aminocaproic acid and tranexamic acid, 50,000 KIU /Kg of aprotinin and 5.0 mg/Kg of gabexate mesilate before the exposure of cold-restraint stress significantly reduced the severity of gastric ulcerations and hemorrhage These doses of agents did not affect total acid output in pylorus-ligated rats. These results support a concept that thrombogenesis, fibrinolysis and kinin system in the gastric mucosa might play a role in the pathogenesis of stress ulcers and that these agents could be applicable for clinical use.
