MONOCLONAL ANTIBODIES AGAINST CARCINOMA OF THE STOMACH AND COLON FOR DIAGNOSIS AND TARGETING CHEMOTHERAPY

Monoclonal antibodies against scirrhous carcinoma of the stomach and carcinoma of the colon were produced from hybridomas of mouse myeloma cells and sensitized mouse spleen cells.
Monoclonal antibodies (S43, S74, S202) against scirrhous carcinoma of the stomach responded more than 90 percent to carcinoma of the stomach, being suggestive of diagnostic value.
Monolonal antibody (A7) against carcinoma of the colon responded specifically 52 percent to carcinoma of the colon. 125I-leveled A7 was injected into the nude mice with transplanted human colon cancer to evaluate localization of antibody. Antibody was localized selectively at the subcutaneously transplanted tumor.
Monoclonal antibody was covalently bound to Mitomycin C (MMC) and Neocarcinostacin (NCS), and A7-MMC and A7-NCS were produced for targeting chemotherapy. A7-MMC and A7-NCS remained both activities of antibody and anticancer. A7-MMC had 100 times cytotoxic effceffect on SW-1116 cells, as compared with conventional MMC. A7-MMC and A7-NCS inhibited growth of colon cancer in nude mice.
A7-NCS was injected into 23 patients with colon cancer, 6 patients with liver metastasis of colon cancer, 1patient with lung metastasis and peritoneal metastasis of colon cancer respoectively. A7-NCS was detected in resected colon cancer. Five out of 6 patients with liver metastasis responded favorably to A7-NCS. Sixteen patients had fever for one to three days after injection, but no serious side effect was noticed.
