The proximal tubules (PT) of the dog kidneys were examined by transitional electron microscope after various warm ischemia time (WIT) and cold ischemia time (CIT). Lactated Ringer (RG) or modified Collins’ solution was used for simple cold preservation, and LR or cryoprecipitated fibrinogenfree plasma (CFP) for cold perfusion preservation.
The following results were obtained.
1) The cells were in good preservation at 30 min. of WIT, but the necrotic degeneration appeared at 60 and 90 min. of WIT.
2) The vacuolization of the cells progressed in parallel with the duration of WIT.
3) In the simple cold preservation with Collins’ solution, the damages of cells were less than with LR.
4) In the perfusion preservation with CFP, the swelling of cells was comparatively less at early stage but severe changes of cells developed after 8 hours.
5) In the perfusion preservation with LR, selective mitochondrial damage was recognized.
It was concluded that the morphological changes of cells developed rapidly after 30 min. of WIT and the simple cold preservation with Collins’ solution was most excellent at least within 10 hours.
