THE IMPAIRED CARBOHYDRATE METABOLISM IN ENDOTOXEMIA EXPERIMENTAL STUDY BY THE IN VIVO AND PERFUSED RAT LIVER
The levels of several hepatic glycolytic intermediates were studied in vivo and in perfused rat liver, with or without endotoxin. At four and eight hours after the administration of endotoxin, there appeared marked hypoglycemia and depletion of glycogen, hexosephosphate and phosphoenolpyruvate (PEP) in the liver, while fructosediphosphate (FDP) and lactate were elevated.
In perfusion study for 60 minutes with endotoxin in well oxygenated condition, PEP and following gluconeogenic metabolites were found to be reduced, but the elevation of FDP and lactate were not observed. On the other hand, increased levels of FDP and lactate were shown by perfusion study in hypoxic condition. The reduction of glucose production was seen by either way of endotoxin ariministration or hypoxic condition in perfused liver.
It is concluded that the most likely cause of decreased gluconeogenesis in endotoxemia suggested by these results was an inhibition of some gluconeogenic enzymes (e.g. PEPCK) by endotoxin itself, in addition to the liver cell hypoxia.
