STUDIES ON ESTROGEN RECEPTORS IN HUMAN BREAST CANCER BY GEL-FILTRATION METHOD
Estrogen receptors were measured by using gel-filtration method in cytosols from human breast cancers, DMBA induced rat mammary tumors, rabbit uteri, rat uteri and various human organs.Sixty percent inhibition by nonradioactive estradiol-17β and fifteen femtomoles per mg of cytosol protein were available critical values between receptor positive case and negative one. The former revealed to be equal or over the above levels and the latter was always under them. In addition there was a intermediate borderline combination case. This criteria of estrogen receptors showed good correlations to the biological estrogen dependency in the DMBA induced mammary tumors, rabbit uteri, rat uteri and various human organs. Estrogen receptors were found to be positive in 45.3% among 86 breast cancers. There was no significant correlation between the presence of the receptor and some clinical factors such as, clinical stage of cancer, tumor size, number of axillary lymphnode metastasis,histological type, menopausal status, or urinary steroids excretion. Primary and metastatic lesions which obtained from the same patient simultaneously or sequentially showed no significant discrepancies in the results of estrogen receptors. In metastatic or advanced breast cancers, 7 out of 9 patients with receptor-positive tumors had objective regressions to endocrine treatment. None of 13 patients with receptor-negative tumors had objective regressions.
The measurement of estrogen receptors by using gel-filtration method was thought to be useful to predict the effect of endocrine treatment in patients with breast cancer.
