The activity of cholinesterase (Ch-E) in plasma of blood in circulation and in the mesenteric and intestinal tissues was determined by Ammons method in cats in experimental ileus and in normal cats for comparison. The following results were obtained.
1. The activity of Ch-E in plasma of blood in systemic circulation was slightly below normal for two days following simple laparotomy and brought up to normal later in control animals.
2. It was conspicuously decreased in course of time in animals in ileus, change in level of the activity not markedly depending on the particular part of the intestine at which ileus was caused.
3. It was kept at a still lower level for some time after removal of the obstruction, .and it took a week or more to be restored to its original level.
4. The activity of Ch-E in plasma in blood of a mesenterial vein was not markedly varied in intensity in different parts of the small intestine when the intestine was in a normal state, but when the intestine was obstructed, it was greater at a point immediately :above the obstructed part than at any point further a part from it.
5. The activity of the esterase in tissue of the small intestine was varied by degrees, .and not uniform, in intensity in different parts of the tissue: it was greater in the lower than in the upper part.
6. It was generally below normal when the int stin was obstructed: it was invariably smaller then at a point just above the obstructed part of the intestine than at a point just below it, becoming more markedly so at the former point with the duration of obstruction.
