EXPERIMENTAL AND CLINICAL STUDIES ON NITROGEN MUSTARD
The author used Tris (β-chloroethyl) amine (abbreviated NH
3) and Methyl bis (β-chloroethyl) amine (abbreviated NH
2) of Nitrogen Mustard (abbreviated N.M.) since the Spring of 1948, and obtained the following results ;
Animal experiment. 1. Having pursued change of Yoshida sarcoma cell, the author came to ascertain that N.M. acts on the cell to bring a remarkable regressive degeneration and most strongly especially on the cells which are in the course of mitosis.
2. As the result of comparison and investigation of the sensibility of the cells usually exsting in the rat abdominal cavity and the cells of Yosida sarcoma through percentage in 500 cells, a selective effect was seen only in the sarcoma cells by per kg of weight 0.1 mg of N.M.
3. As the result of injections performed at each period directly after the transplantation of Yosida sarcoma and Ist, 4th and 9th day after the transplantation, the author recognized that a extremely remarkable effect appeard directly after the transplantation was over, and that subsequently the effect decreased with the lapse of time after the transplatation.
4. As the result of comparison between injection into the vein and into the abdominal cavity perfomed against Yosida sarcoma through percentage of mitosis in the neoplastic cells was known that direct injection into the abdominal cavity is far more effective than that into the vein.
5. It is obvious that N.M. resolves to decrease its effect after dissolving. The author has searched for hourly changes of it numerically, using mouse lethal dosis and changes of effect by the treatment of Yosida sarcoma and recognized that though N.M. should be used swiftly to apply clinically after its dissolution, there is no necessity to determine the period by odd minutes or seconds, because defference is slight experimentally so long as it is used within 24th hour after its dissolution.
6. As the result of comparison between NH
3 and NH
2 used for the purpose of healing Yosida sarcoma, much more difference between toxication dosis and healing dosis was seen in NH
2 than NH
3 and the healing result of the latter was better, and a modicum and continuity treatment as means of healing was most available.
7. As the result of a histological observation of influence on the healing of the incised wounad by means of cutting the skin of rabbit and injecting 0.2 mg of N.M. per kg at the same time, it was known that there happens no malignant influence.
8. The author iniected the lethal dosis of N.M. into mouse and also various medicine at the same time in order to find anything able to decrease the lethal dosis, but none of them showed any effect.
