STUDIES ON DIFFERENTIATION OF CELLS ORIGINATED FROM HUMAN NEURAL CREST TUMORS IN VITRO, IN VIVO AND IN CLINICAL CASES
Artificial induction of differentiation of human neural crest tumors was studied using various kinds of chemicals in vitro, in vivo and in clinical cases. DBcAMP, cholera toxin, N-butyric acid, dopamine hydrochloride, Con A, serum free medium, K-247, mitomycin, and ascites were found to enhance the extension of dendrite-like processes of human neuroblastoma cells (NB-1) and human melanoma cells (TM-1) in vitro. DBcGMP, dexamethasone, galactose, vitamine C, X-ray and culture of low temperature enhanced DOPA reaction, but did not stimulate the cellular processes. By Papanicolaou staining, keratinization of well differentiated squamous cell carcinoma (TE-1 cells) of the esophagus was occurred without any special treatment for cells. Treating with 1 mM DBcAMP combined with vitamine A, the cytoplasma : nucleus ratio was enlarged and the shape of TE-1 became similar to the normal cultured cells of the esophagus.
Regression of human tumors transplanted to nude mice was studied by injecting DBcAMP and theophylline intraperitoneally or cholera toxin into tumors. Prolongation of survival time of nude mice was found in these treated animals. 4 year and 6 month old girl (Stage IV neuroblastoma) given palliative operation was treated with 50 mg neophylline, i.v., 100 mg trental, P.O., and maximally 1000 mg/day DBcAMP. HVA/VMA ratio was markedly decreased soon after the operation and gradually done after the treatment of the above agents. Slight nausea and remarkable tachycardia were noted as side effects of a large amount of DBcAMP.
